Abstract # 1049 Histone Deacetylase Inhibitor LBH589 Radiosensitizes Non-Small Cell Lung Cancer by Inhibiting DNA Repair

H23 and H460 NSCLC cell lines were used for all studies. Clonogenic survival assays were performed with LBH589 treatment prior to 0-6 Gy. Apoptosis was measured using Annexin-PI flow cytometry assays, DAPI staining for nuclear morphology, and cleaved caspase 3 Western blot analysis. An in vivo tumor growth delay assay was performed using a hind limb xenograph model with two oral treatments of 40 mg LBH589 and five 2 Gy fractions over seven days. To study the effect of LBH589 on radiation induced DNA double-strand break repair immunostaining was performed for γ-H2AX nuclear foci. Furthermore, the compartmentalization of HDAC4 was studied using immunostaining and Western blot analysis. Student's t-test was used to determine statistical significance.
